This test method describes the enzyme-substrate-based detection of total coliforms and Escherichia coli in drinking water, surface water, and wastewater using the Colilert® test. This method is based on defined substrate technology, where specific enzymes produced by coliforms and E. coli react with added substrates to produce color and fluorescence, allowing for easy detection and enumeration after an 18-hour incubation period.
